{"id":462,"date":"2021-01-06T17:15:44","date_gmt":"2021-01-06T16:15:44","guid":{"rendered":"https:\/\/staging.blogs.yokogawa.de\/life-innovation\/2021\/01\/06\/high-content-imaging-for-3d-spheroid\/"},"modified":"2022-07-21T11:46:14","modified_gmt":"2022-07-21T09:46:14","slug":"high-content-imaging-for-3d-spheroid","status":"publish","type":"post","link":"https:\/\/www.yokogawa.com\/eu\/blog\/life-innovation\/high-content-imaging-for-3d-spheroid\/","title":{"rendered":"High Content Imaging for 3D Spheroid"},"content":{"rendered":"<p>We would like to invite members of the drug\u00a0discovery\u00a0community to learn about special considerations for\u00a03D\u00a0Spheroid\u00a0and Organoid\u00a0Imaging.\u00a0 Together with experts at\u00a0InSphero\u00a0and Yokogawa,\u00a0 we are hosting a webinar on January 14, 2021.\u00a0<strong>In this webinar,\u00a0 we share our experience and\u00a0<\/strong><strong>advice for sample preparation, imaging, and analysis for both fixed and live-cell in high content screening assays. <\/strong>We hope to provide the\u00a0research\u00a0community with tips and tricks\u00a0for 3D drug screening,\u00a0highlighting\u00a0practical aspects\u00a0of\u00a0drug screening\u00a0in\u00a0discovery and therapy\u00a0with 3d\u00a0multicellular\u00a0models.\u00a0\u00a0<\/p>\n<h2><strong>High content imaging for drug screening<\/strong><\/h2>\n<p>Conventional drug screening has mostly been limited to 2d cell cultures in multi-well plates.\u00a0The 2D cell culture model is being replaced with 3D models as a more physiologically relevant in vitro model choice.\u00a0The behavior of cells differs in three-dimensional cell contacts. Tissue relevant co-cultures and integrated signaling, along with o2\/co2, nutrient, waste, and drug gradients are\u00a0quite different\u00a0in 2D vs 3d. High content imaging enables clinically relevant interrogation of 3d models, along with targeting of specific cell-cell interactions of target and non-target cell types.\u00a0Using spinning disk confocal microscopy allows long-term imaging of live samples due to low phototoxicity and optical sectioning over 100um deep into 3d tissue samples. \u00a0<\/p>\n<p>With\u00a0careful choice of instruments and sample holders,\u00a0even month-long longitudinal studies of live samples are possible.\u00a0\u00a0We will show some examples of\u00a0how\u00a0live imaging vs fixed imaging can change\u00a0regarding\u00a0throughput, kinetic data,\u00a0probes,\u00a0and signal strength.\u00a0\u00a0<\/p>\n<p><a href=\"https:\/\/www.labroots.com\/ms\/webinar\/optimizing-high-content-imaging-3d-models-drug-discovery-2?utm_source=blog&amp;utm_medium=banner&amp;utm_campaign=yca_webinar_en\" target=\"_blank\" rel=\"noopener\"><img decoding=\"async\" class=\"aligncenter wp-image-1097 size-full\" src=\"https:\/\/www.yokogawa.com\/eu\/blog\/app\/uploads\/sites\/9\/2022\/06\/banner.png\" alt=\"high content imaging webinar\" width=\"452\" height=\"237\" \/><\/a><\/p>\n<h2><strong>Looking at cell level data<\/strong><\/h2>\n<p><img decoding=\"async\" class=\"alignright wp-image-1104 size-medium\" src=\"https:\/\/www.yokogawa.com\/eu\/blog\/app\/uploads\/sites\/9\/2022\/06\/drug-screening-300x254.jpg\" alt=\"high content imaging\" width=\"300\" height=\"254\" \/>It is important to keep in mind:\u00a0 probe and antibody penetration kinetics, taking into consideration\u00a0sample\u00a0fixation\u00a0and permeabilization conditions, along with incubation time, and any non-specific binding that may occur.\u00a0For fixed tissue, tissue clearing can be\u00a0an excellent choice to increase light penetration allowing for a larger sample size, which can be important to understand the larger picture of cell-cell interactions.\u00a0By looking at cell level data, we can count cells, look for intracellular biomarker expression, individual cell fate populations, along any intercellular structures. The spatial information along with cell infiltration and migration or signaling in defined zones due to kinetic changes in cell number or biomarker expression can be achieved in live sample preparation.\u00a0\u00a0<\/p>\n<h2><strong>How to understand the complex volume of data<\/strong><\/h2>\n<p>We will discuss common ways to report results of analysis, including heatmaps, dose-response curves, and per well pie charts.\u00a0We\u00a0will include a brief\u00a0discussion\u00a0of Machine Learning and Deep learning tools for segmentation and phenotypic assignment. These automated tools can help researchers to understand the complex volume of data in these high content imaging analysis systems.\u00a0<\/p>\n<p>I\u2019m looking forward to an exciting discussion.
