Subcellular Sampling System
SS2000은 배양 중인 세포를 공초점 현미경으로 이미징하면서 단일 세포 수준에서 세포의 특정 영역 또는 세포 전체 영역을 자동으로 샘플링하는 시스템입니다. 이는 배양 중인 세포를 분리할 필요가 없기 때문에 셀의 위치 정보 및 형태학적(morphological) 정보를 유지시킬 수 있습니다.

Use Cases
- 단일 세포 수준에서 세포 내 성분을 샘플링할 수 있습니다. 여기에는 지질막이 없는 소기관(organelles without lipid membranes)과 같이 생화학적 방법으로 샘플링하기 어려운 세포내 구성요소도 포함됩니다.
- 위치정보를 유지하면서 샘플링함으로써 암세포에 인접한 정상세포와 암세포에서 멀리 떨어진 정상세포를 샘플링하고 분석하는 것이 가능합니다.
- 형태학적 정보(morphological information)를 유지하면서 샘플링이 가능합니다. 다양한 형태학적 변화를 가진 세포를 샘플링하고 비교할 수 있습니다.
- 세포체나 axon과 같은 뉴런(neuron)의 다양한 부분을 샘플링 대상으로 삼을 수 있습니다.

Subcellular sampling
SS2000은 기존 세포 분리 장치와 다르게 전체 세포를 분리할 수 있을 뿐만 아니라 세포 내부의 표적 부위만도 샘플링 할 수 있습니다. 이는 세포질(cytoplasm)과 표적 소기관(target organelles)이 포함된 영역을 선택적으로 샘플링 할 수 있다는 것을 뜻합니다.

Maintaining positional and morphological information of cells
배양 중인 세포를 분리하지 않고 표적 세포만 채취할 수 있어 위치 및 형태학적 정보를 유지한 채로 채취가 가능합니다.
정상 MDCK 세포와 녹색 형광으로 표지된(labeled) 비정상 MDCK 세포를 50:1 비율로 공동 배양하였으며 형광 신호(화살표)를 나타내는 비정상 세포에 인접한 정상 세포를 샘플링.
High usability samples
샘플은 PCR 플레이트 및 마이크로 플레이트에서 수집할 수 있으며 동일한 웰에서 여러 샘플을 수집할 수도 있습니다. 샘플 ejection 없이 유리 팁 안에 있는 채로 샘플을 채취할 수 있습니다. 샘플 collection 위치에는 검체 분해를 억제하는 냉각 기능과 배양 환경을 유지하는 인큐베이터 기능이 포함되어 있습니다. 이러한 샘플은 유전자 분석, 질량 분석 및 단일 세포 복제(single-cell cloning)에 사용될 수 있습니다.
Live Cell Imaging with Confocal Microscopy
SS2000은 Yokogawa가 자체 개발한 라이브 셀 이미징 장치를 사용합니다. 당사 고유의 공초점 현미경 기술을 사용하여 고속, 고해상도 3D 이미징이 가능합니다. 인큐베이터 환경에서 공초점 현미경을 이용하여 표적 세포로부터 샘플을 채취할 수 있습니다. 시간 경과 촬영(Time-lapse photography)도 가능하므로 대상 셀의 동적 변화도 캡처할 수 있습니다. 샘플링 중, 샘플링 전후의 영상도 동영상으로 기록할 수 있기 때문에 수집된 시료의 분석 결과를 세포 영상 데이터와 비교할 수 있습니다.
이미지 분석을 통해 타겟 셀과 그 샘플링 위치를 자동으로 선택할 수 있습니다. (세포의 모양, 핵의 크기, 세포 소기관(organelles)의 밀도 등으로 대상을 자동으로 선택할 수 있습니다)
