Synopsis

Process Analytical Technologies (PAT) have become a powerful tool of biopharmaceutical organizations to monitor and control Critical Quality Attributes (CQA) of manufactured biologic drugs.  Effective implementation of upstream PAT by employing spectroscopic techniques is a challenging endeavour requiring accurate cell models and algorithms to stabilize culture conditions, especially with regard to nutrients.   With a unique cell modelling design approach and model predictive control (MPC) software, we have created a robust fully-integrated lab-scale bioreactor system that is adaptable and provides precision glucose feed control.  Using multiple bioreactors in parallel controlled with a laboratory process information management system, we have demonstrated operational consistency and the utility of this approach for exploring optimal glucose requirements of antibody-producing CHO cells in cell culture.
