Cell Cycle

In this tutorial, we will identify the cell cycles G1-phase, G2/M-phase, etc. using the intranuclear DNA content. DNA content is measured from the fluorescence intensity of images obtained using Hoechst staining. If we make the DNA content per nucleus equal to “1” for the G1-phase, then the DNA content doubles and becomes [2] for the G2-phase, due to the occurrence of DNA synthesis in the S-phase. Using this property, cycles are classified using the aggregate amount of intranuclear fluorescence intensity following nucleus recognition.

For the analysis, the [CellCycle] protocol pre-installed in [Application] will be used to recognize the nuclei, and the cell cycles G1-phase, G2/M-phase etc. will be classified using the Gate function. Following this, the proportion of each -phase will be displayed using a pie chart.
